Abstract

The aim of this work was to evaluate in vitro organogenesis as a function of growth regulators and to test the influence of 2,4-D inducing somatic embryogenesis in basilhybrid ‘Sweet Dani’ x ‘Maria Bonita’. We used the completely randomized design in the trials. In the first experiment, we used a 6x4 factorial scheme, testing six concentrations of BAP and four concentrations of IAA. In the second experiment, we tested four types of culture medium, and in the third assay, we evaluated five concentrations of BAP. In the fourth trial, we evaluated the effect of seven concentrations of BAP combined with 0.20 mg.L-1of NAA. The fifth experiment was conducted in a 5x2x2factorial scheme, testing five concentrations of 2,4-D, two culture media and two leaf positions. The absence of BAP and IAA, in the culture medium, resulted in a greater number of leaves and shoots, and higher fresh and dry weight of explantshoots. The shoot regeneration via direct organogenesis occurred when explants were maintain in medium with 0.8 mg.L-1of BAP, and indirect organogenesis with 1.9 mg.L-1of BAP. 2,4-D provided the formation of calluses with embryogenic characteristics in the tested media cultures, independently of leaf position. Concentrations of 0.5 and 1.0 mg.L-1of 2,4-D in MS and MT culture media, with leaf in abaxial position, showed a higher number of embryogenic cells and formation of globular embryos.
